反义α肌动蛋白基因腺病毒载体转染5/6肾切除大鼠抑制肾纤维化进展的研究

摘要 目的观察大鼠血管平滑肌α肌动蛋白基因反义腺病毒载体(pAdanti-α-SMA)转染5/6肾切除大鼠对其肾脏纤维化进程的影响。方法建立pAdanti-α-SMA转染5/6肾切除大鼠模型。实验分组,A组:正常对照;B组:5/6肾切除大鼠;C组:pAdanti-α-SMA转染的5/6肾切除大鼠。统计20周后存活率及血肌酐,行肾脏病理检查和肾脏损伤指数评分,行α-SMA、TGF-β1、PDGF-BB的免疫组化并用Tiger920图像分析仪做半定量分析。结果各组存活率为95.0%、58.8%、82.9%;各组平均血肌酐为:39.74、132.57、88.65;各组肾损伤总指数:0.79、16.91、1.43;各组α-SMA+细胞Tiger920图像分析仪结果:1.22、1.81、1.37。结论pAdanti-α-SMA转染可延缓大鼠肾脏纤维化进程。 Objective To evaluate the inhibiting of renal fibrosis progression of 5/6 nephrectomized rats transfected by Transgene of Antisense Vascular Smooth Muscle Alpha Actin(pAdanti-α-SMA). Methods To transfect 5/6 nephrectomized rats by the pAdanti-α-SMA. Rats survival rate,weight, renal function and renal tissues pathology were assessed. Expression of α-SMA,TGF-β1 and PDGF-BB in kidney were detected by immunohitochemistry and analyzed by Tiger 920. Results 5/6 nephrectomized rats were successfully transfected by pAdanti-α-SMA. The survival rate of 5/6 nephrectomized rats transfected by pAdanti-α-SMA was higher than that of rats which were not transfected. The renal function and renal tissues pathology of the transfected rats was better than that of rats which were not transfected. Quantity of α-SMA+ cells in rats transfected was higher than that in rats which were not transfected ,while the quantity of the TGF-β1 + and PDGF-BB+ cells both showed no difference in the same experiment. Conclusion The RFB progression of 5/6 nephrectomized rats transfected by pAdanti-α-SMA is inhibited.
