茶树幼胚培养萌发率与再生途径影响因素研究 被引量:10

摘要 从激素浓度与组合、基本培养基和蔗糖浓度等几个方面进行了茶树幼胚培养中萌发率和再生途径的影响因素研究。试验结果表明,茶树幼胚的萌发需要有一定量的生长素的剌激作用,但较强烈的细胞分裂素的作用对茶树幼胚的萌发有抑制作用;所使用的激素种类对于胚的再生途径有较大的影响,在MS+KT2.0+IBA0.1+GA31.0mg/L培养基上,80%以上的幼胚可以继续发育成熟并萌发成苗;而高糖浓度则会诱导产生较多的胚性愈伤组织并从而导致不定胚的形成。 The effects of different kinds of basic medium,levels of sucrose and the level as well as the combination of phytohormones on the germination rate and the regeneration pathway of tea young embryos incubated were investigated. The result indicated that the germination of young embryos needed the stimulative effect of auxin in low level, but the high level of cytokinin would restrain the germination. The kinds of hormone used had biggish influence on the regeneration pathway employed by the incubated young embryos. Over 80 % of young embryos could continue developing to mature and sprout to form seedlings on the media of MS + KT 2. 0 + IBA 0. 1 + GA3 1.0 mg/L. The result also indicated that higher level of sucrose would result much more embryonie callus and then induce formation of adventive embryos.
