摘要
目的探讨铜绿假单胞菌(Pseudomonas aeruginosa,PA)感染与人巨噬细胞系U937细胞凋亡的关系。方法以U937细胞作为PA感染的体外细胞模型,使细胞与细菌浓度比分别为1∶10、1∶20、1∶50及1∶100,用荧光染色技术、Annexin V/PI双染流式细胞仪检测细胞凋亡率。结果细胞与细菌比例为1∶10时即可引起部分U937细胞发生凋亡,Hoechst染色法和Annexin-V FITC/PI双染流式细胞仪检测细胞凋亡率分别为(11.67±1.75)%和(11.68±2.58)%,且U937细胞凋亡百分率随PA感染剂量增加而增加。结论铜绿假单胞菌感染可诱导U937细胞凋亡,且呈剂量依赖。
Objective To investigate the relationship between Pseudomonas aeruginosa(PA) and the apoptosis of U937 cell. Methods U937 cell apoptosis was induced by PA in groups of U937: PA= 1∶10,1∶20,1∶50 and 1∶100,respectively,and detected with Hochest 33258 fluorochrome staining and Annexin V/PI assay. Results When the ratio between cell and bacterium was 1∶10,the apoptotic cells were found in all the groups.Hoechst 33258 fluorochrome staining and Annexin V/ PI assay showed that the percentage of apoptotic U937 for(11.67±1.75)% and(11.68±2.58)%,and the percentage of apoptotic cells was raised with the increase of the concentration of PA.Conclusion PA can induce the U937 cells apoplosis in a dose-dependent manner.
