分子荧光标准曲线法对不同龄兔晶状体摘除术后血-房水屏障破坏的动态观测 被引量:1

摘要 目的探讨幼年和成年兔血一房水屏障存在的差异和晶状体摘除术对此屏障的影响。设计随机对照实验研究。研究对象新西兰白兔,幼年和成年各10只。方法随机选取健康幼年和成年新西兰白兔各10只,摘除右眼晶状体,分别在手术前7天和术后1、3、7、14天自右侧耳缘静脉按30 mg/kg体重(0.15 ml/kg体重)快速注射20%荧光素钠注射液,60分钟后抽取房水,用分子荧光标准曲线法测定手术前7天左眼(未手术)和右眼术后1、3、7、14天房水荧光素浓度,记录两组兔生理状态和各组手术前后不同时间点的房水荧光素浓度,并对结果进行比较。主要指标房水荧光素浓度。结果生理状态下幼兔房水荧光素浓度明显高于成年兔(P=0.015),分别为(617.95±178.29)μg/L和(427.39±135.23)μg/L;术后两组兔荧光素渗漏均明显增加,3天时达到高峰,分别为(3070.05±1083.22)μg/L和(3854.03±1059.67)μg/L;14天时两组兔房水荧光素含量和术前相比没有统计学差异,分别为(1057.72±284.49)μg/L和(1110.07±339.54)μg/L,P值分别为0.086和0.125。结论幼兔血-房水屏障的通透性较成年兔高,屏障功能发育不完善;晶状体摘除手术会造成兔眼血-房水屏障功能损害。(眼科,2008,17:116-119) Objective To investigate the differences of blood-aqueous barriers between young and adult rabbits and the influence of surgery after extra-capsular lens extraction. Design Randomized, controlled experimental study. Participant Different aged New Zealand rabbits, comprise 10 young and 10 adult ones. Method 10 young and 10 adult New Zealand rabbits were selected at random, and extra-capsular lens extraction was performed on the right eye of each rabbit. 20% Fluorescein Sodium was quickly injected into the ear-vein with the dosage of 30 mg per kilogram body weight on 7d before surgery and 1, 3, 7 and 14d after surgery. 60 minutes later at each time point, aqueous humor was extracted, and Molecule-fluorescence-standard curve method was applied to test the concentration of fluorescence in anterior chamber at above time points. The concentration of fluorescence under the physiologic condition between young and adult was compared. They were also been compared respectively with those at different time points after surgery. The results were analyzed with the statistic software SPSS 11.5. Main Outcome Measure The concentration of fluorescence in anterior chamber. Result Physiologically, the concentrations of fluorescence in anterior chamber were more higher in young rabbits than in adult ones, the former was 617.95 ± 178.29 μg/L, and the latter was 427.39 ± 135.23 μg/L(P=0.015). The fluorescence leakage was obviously increased after surgery, and the peak time point was the 3rd postoperative day, 3070.05±1083.22 μg/L in young group and 3854.03±1059.67μg/L in adult group. The contents of fluorescence at pre-operative day and at 14th post-operative day were compared respectively in two groups. They were 1057.72 ± 284.49 μg/L and 1110.07 ± 339.54 μg/L, and no significant differences was found (P=0.086 in young and P=-0.125 in adult). Conclusion In young rabbits, the blood-aqueous barrier is unsound and its permeability is abnormally increased. The function of the blood-aqueous bairier was damaged after the surgery.
