摘要
目的探讨转化生长因子β1(TGFβ-1)对大鼠心脏移植排斥反应中CD44、选择素E、淋巴细胞功能相关抗原-1(LFA-1)、血管细胞黏附分子-1(VCAM-1)表达的影响。方法采用大鼠颈部心脏移植模型,随机分为3组:对照组、移植组、TGF-β1组。于移植术后第5天取移植心脏,用逆转录聚合酶链式反应(RT-PCR)法观察CD44、选择素E、VCAM-1、LFA-1的表达情况。结果移植组CD44、选择素E、LFA-1、VCAM-1表达明显升高,与对照组相比,均P<0.01。与移植组相比,TGFβ-1组CD44、选择素E、LFA-1、VCAM-1表达明显降低,均P<0.01。结论TGF-β1对心脏移植排斥反应的免疫抑制作用可能与其抑制CD44、选择素E、LFA-1、VCAM-1等黏附分子的表达有关。
Objective To study the effect of TGF-β1 on the expression of CD44, selectin-E, LFA-1 and VCAM-1 in rat heart transplantation rejection. Methods The model of rat cervical heterotopic heart transplantation was established. The rats were divided into control group, transplant group and TGF-β1 group. The expression levels of CD44, selectin-E, LFA-1 and VCAM-1 mRNA was detected by using RT-PCR at 5th day after transplantation. Results The expression levels of CD44, selectin-E, LFA-1 and VCAM-1 mRNA were significantly increased in transplant group as compared with control group (all P〈0.01). In TGF-β1 group, the expression levels of CD44, selectin-E, LFA-1 and VCAM-1 mRNA were significantly decreased as compared with transplant group (all P〈0.01). Conclusion TGF-β1 may decrease the expression of CD44, selectin-E, LFA-1 and VCAM-1 in rat heart transplantation rejection and prolong the survival days of transplanted hearts.
