血清胰岛素样生长因子-1水平对癌性恶病质小鼠营养状况的影响 被引量:1

Effect of Serum IGF-1 Level on Nutritional Status of Cancer Cachexia Mice

摘要 目的探讨血清中胰岛素样生长因子-1(IGF-1)水平与恶病质小鼠营养代谢的关系。方法肝脏特异性IGF-1基因缺失鼠(LID鼠)和对照组小鼠各20只接受皮下接种小鼠结肠腺癌CT26细胞株,建立癌性恶病质模型。两组小鼠分别又随机分成2个亚组:对照组10只、LID组10只接受皮下注射重组人生长激素(rhGH,1 mg/kg),每天1次,从肿瘤种植后第14天开始至第22天实验结束;其余小鼠接受皮下生理盐水注射至实验结束。统计各组小鼠进食量、肿瘤大小和重量、除瘤后体重、腓肠肌重、腓肠肌总蛋白量、血清中白细胞介素-6(IL-6)及肿瘤坏死因子-α(TNFα-)水平。结果所有接种动物均出现恶病质。接受rhGH注射的LID鼠和对照组小鼠血清IGF-1水平均明显升高(P<0.05)。血清IGF-1水平较高的小鼠与血清IGF-1水平较低的小鼠比较,腓肠肌重和腓肠肌总蛋白量均高于后者(P<0.05),血清中IL-6及TNF-α水平低于后者(P<0.05),肿瘤大小和进食量均无明显差异(P>0.05)。结论从恶病质早期开始进行外源性的GH注射可引起恶病质小鼠血清IGF-1水平的明显升高,并可以改善癌性恶病质机体骨骼肌蛋白代谢,缓解恶病质症状。IL-6和TNFα-水平与恶病质机体营养代谢相关。 Objective To detect the relationship between the serum insulin-like growth factor 1 (IGF-1) level and nutritional status of mice with cancer cachexia. Methods Colon 26-adenocarcinoma tissue fragments were transplanted subcutaneously into the liver-specific IGF-1-deficient (LID) mice (n=20) and control mice (n = 20). The LID mice and control mice were randomly divided into two subgroups respectively, with 10 LID mice and 10 control mice being injected subcutaneously with rhGH (1 mg/kg) once daily from the 14th day after transplants to the 22nd day and the remaining mice receiving saline injection. The food intake, body weight, tumor size, the weight and total protein level in gastrocnemius muscle were documented. Serum interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α) levels were also analyzed. Results The treatment of GH increased the serum levels of IGF-1 in both LID mice and control mice (P〈0.05). The gastrocnemius muscle weight was heavier and total protein level higher in the mice with higher serum IGF-1 levels accompanied by decreased serum IL-6 and TNF-α levels than those with lower serum IGF-1 levels (P〈0.05). There was no statistically significant difference in the food intake and tumor size (P〉0.05). Conclusion The treatment of GH from the early period of cancer cachexia could increase serum IGF-1 levels and the lean body mass significantly. Serum IL-6 and TNF-α may be partially attributed to the catabolism of skeletal muscle and so are correlated to the cancer cachexia.
