摘要 目的探讨Toll样受体4(Toll-like receptor 4,TLR4)的表达与脑组织缺血再灌注损伤的关系。方法线栓法制备局灶性脑缺血再灌注损伤大鼠模型,采用免疫组织化学法观察TLR4在不同再灌注时间点缺血侧脑组织的分布和表达,同时应用酶联免疫吸附方法检测白细胞介素1β(interleukin-1β,IL-1β)在不同再灌注时间点缺血脑组织内的浓度。结果大鼠局灶性脑缺血再灌注损伤过程中,TLR4蛋白在皮质缺血半影区及邻近纹状体区有表达,且在再灌注后1 h即达高峰;IL-1β在再灌注后3 h明显升高[(4.47±2.13)ng/ml],表达高峰在再灌注后12 h[(14.39±2.58)ng/ml]。结论脑组织缺血再灌注过程中TLR4蛋白的表达有可能作为重要炎性受体介导脑缺血炎性损伤。 Objective To explore the relation between the expression of TLR4 and the partial cerebral ischemia/reperfusion (I/R) injury in rats. Methods Thread embolism was performed to establish partial cerebral I/R model in rats. The expression and distribution of TLR4 in ischemic cerebral tissues were determined by immunohistochemistry and the concentration of IL-1β in ischemic cerebral tissues was assayed by ELISA at different reperfusion time points. Results During the process of partial cerebral I/R injury in rats, the expression of TLR4 protein was increased and reached the peak at 1 h after reperfusion in the cerebral cortex of the ischemic penumbral region and corpus striatum. The level of IL-1β was increased gradually after reperfusion for 3 h [(4.47±2.13) ng/ml] and reached the peak at 12 h after reperfusion [(14.39±2.58) ng/ml]. Conclusion The expression of TLR4 may mediate the inflammatory injury in the process of partial cerebral I/R as one of important inflammatory receptors.
