自拟化瘀胶囊对大鼠脑缺血再灌注损伤的保护作用

PROTECTIVE EFFECT OF SELF-MADE HUAYU CAPSULE AGAIST CEREBRAL ISCHEMIA-REPERFUSION INJURY IN RATS

目的:观察化瘀胶囊对大鼠脑缺血再灌注损伤的保护作用。方法:Wistar大鼠雄性60只,随机分为6组,每组10只。即化瘀胶囊高、中、低剂量组、尼莫地平组、模型对照组和假手术组分别给等体积生理盐水,各组均连续灌胃给药7天,末次给药30min后。参照文献法制备大鼠脑缺血再灌注模型,通过观察模型大鼠脑含水量、脑组织SOD活性、MDA含量及NO含量的变化,探讨中药化瘀胶囊对局部永久性脑缺血的预防和治疗作用。结果:化瘀胶囊各组能降低脑组织含水量,可抑制脑水肿的发生,对缺血脑组织具有一定保护作用;化瘀胶囊高剂量组能明显提高脑组织SOD活性,减少脑组织MDA生成,中剂量组虽能明显增强SOD活性,但减少MDA生成作用不及低剂量组,提示化瘀胶囊对脑缺血的保护作用与抗自由基生成有关;再灌注2h后,脑组织NO明显升高,而化瘀胶囊高剂量组却可减少其生成,说明化瘀胶囊有抑制再灌注脑损伤引起的NO升高作用。结论:化瘀胶囊对大鼠脑缺血再灌注损伤具有明显的保护作用。

Objective：To investigate the protective effects of Huayu Capsule after cerebral ischemia-reperfusion injury in rats. Methods：Sixty male wistar rats were randomly divided into six groups ,10 rats in each group. Low,middle and high dose of Huayu Capsule treated group, nimodipine treated group, model control group and sham operated group. Every group was given the same dose of normal saline and intraperitoneally administered for 7 days and 30 minuts after the last given medicines,Referencing to the documents,we made the model of cerebral ischemia- reperfusion of rats by investigating the content change of brain tissue water,the SOD activity,the quantity of MDA and nitric oxygen （NO） . We investigated the effects of Huayu capsule preventive and therapeutic effects on partial permanent cerebral ischemic. Results：Removing extravasted Huayu capsule can be able to reduce the brain tissue water content, may suppress the hydro - cephalus the occurrence, has certain protective function to the ischemia brain organization each groupm. High dose of Huayu capsule group could increase the activities of SOD and decrease content of MDA in brain tissue, middle dose of Huayu capsule treated group could increase the activity of SOD, Compared with low dose of Huayu capsule treated group,the content of SOD in middle dose of Huayu capsule treated group was significantly decreased. We found that Huayu capsule has a strong protection,which has relation with anti -free radical. After 2 hours of reperfusicen,content of nitric oxygen （NO） in brain tissue has a significant increase, but the content of nitric oxygen （NO） in high dose of Huayu capsule treated group could was significantly decreased. We found that Huayu capsule has an obvious protective function on cerebral ischemia -reperfusion injury in rats. Conclusion： Huayu Capsule provides a protective effect on cerebral ischemia -reperfusion injury in rats.
