感染异育银鲫补体组分C3·C4表达研究

摘要 [目的]了解补体组分在感染性鱼类疾病中的作用,为鱼类疾病的诊断和治疗提供参考。[方法]采用不同浓度(2×106、4×106CFU/ml)的嗜水气单胞菌感染异育银鲫,利用半定量RT-PCR技术检测感染不同时间后肝脏中补体组分C3和C4的表达量。[结果]感染后4、5 h时,异育银鲫肝脏中C3的表达量均显著高于对照组。在感染后4 h时,不同浓度组之间C3的表达量无显著差异。而在感染后5 h时,感染浓度1组(注射2×106 CFU/ml嗜水气单胞菌)中C3的表达量显著高于感染浓度2组(注射4×106 CFU/ml嗜水气单胞菌)。异育银鲫肝脏中C4的表达水平远低于C3。在感染后5 h时,感染浓度1组C4的表达量显著高于对照组,其余处理组与对照组均无显著差异。[结论]补体组分C3在鱼类免疫防御系统中具有非常重要的意义。 [Objective] The research aimed to understand the role of complement components in infectious fish diseases and provide basis for the diagnosis and treatment of fish diseases. [Method] C. auratus was infected by Aeromonas hydrophila with different concn. (2×10^6,4×10^6 CFU/ml). And the expression amount of complement components C3 and C4 in the liver of C auratus at different time after the infection were detected by semi-quantitative RT-PCR technology. [Result] After being infected for 4 and 5 h, the expression amount of complement component C3 in the liver of C. auratus was significantly higher than that in control group. After being infected for 4 h, the expression amount of complement component C3 had no significant difference among different concn, group. After being infected for 5 h, the expression amount of C3 in the 1st infection concn. group (injecting 2×10^6 CFU/ml A. hydrophila ) was significantly higher than that in the 2nd infection concn, group (injecting 4×10^6 CFU/ml A. hydrophila ). The expression level of C4 in the liver of C. auratus was greatly lower than that of C3. After being infected for 5 h, the expression amount of C4 in the 1st infection concn, group was significantly higher than that in control group and that in other treatment groups had no significant difference with that in control group. [Conclusion] Complement component C3 had a very important significance in the immunity defensive system of fishes.
