摘要
目的研究蒙药娜仁满都拉-11对大鼠种植性胃肿瘤增殖及相关细胞周期蛋白表达的影响;从细胞分子水平探讨该药的抗肿瘤作用机制。方法建立Walker-256种植性胃肿瘤大鼠模型。再将肿瘤鼠随机分为3组:娜仁满都拉-11治疗组670 mg/(kg.d)、阴性对照组(生理盐水组)、阳性对照组(5-FU组)。给药(灌胃)2周后取出瘤组织,测其抑瘤率;免疫组化方法检测种植瘤cyclin D1表达;流式细胞术测定种植瘤细胞周期时相分布情况。结果①与阴性对照组比较,治疗组cyclin D1表达下调(P<0.05);②流式细胞术检测显示:治疗组较阴性对照组G0/G1期细胞比例增高,S期细胞比例下降(P<0.05);③娜仁满都拉-11可抑制鼠种植性胃肿瘤的生长,抑瘤率49%。结论娜仁满都拉-11对大鼠Walker-256种植性胃肿瘤具有一定的抑制作用,通过下调cyclin D1表达,阻断瘤细胞由G1期向S期过渡,从而抑制肿瘤细胞生长,可能是娜仁满都拉-11抗瘤作用机制之一。
Objective To study the effect of Na Ren Man Du La-11 on the proliferation of implanted mice gastric tumor and on the cell cycle protein; To explore the function mechanism of the medicine at cell molecular level. Methods To based mice models of the cell line Walker-256 implanted gastric tumor. We divided mice tumor into three groups:(1)the treatment group of Mongolia medicine Na Ren Man Du La-11 670 mg/( kg·d ) , the group of the negative control( 0.9% NaCl) , the group of positive control ( 5-FU ). 2 weeks later tumor tissues were cut off,we mensurated to depress tumor rate ; The expression of examed implanted tumor cyclin D1 by used immunohistochemically; We examed implanted tumor the cell cycle distributing by flow cytometry. Results (1) Compared with the negative control group, the treatment group had down-regulation of cyclinD1 (P 〈 0.05). (2)To mensurat results by flow cytometry: Compared with the negative control group, the treatment group cell Go/G1 cycle rate is significantly high, cell S cycle down-regulation( P 〈 0.05 ). (3)Na Ren Man Du La-11 could depress the growth of implanted gastric tumor, depress tumor rate 49%. Conclusion Mongolia medicine Na Ren Man Du La-11 can depress the function of the mice cell line Walker-256 implanted gastric tumor; This probably express through down-regulation cyclin D1 ,this probably change cell cycle G1→S1 ,it has some effect on depressing the growth of tumor cells and obviously improve life qualities of the mice with tumor. This probably was one of depressing tumor mechanism of Mongolia medicine Na Ren Man Du-La-11.
