部分纯化小麦β-葡萄糖苷酶的酶学性质

摘要 进行了8个小麦品种中β-葡萄糖苷酶含量的测定,研究发现东农91-5992中β-葡萄糖苷酶含量最高。对东农91-5992中β-葡萄糖苷酶进行了提取、硫酸铵沉淀分离和酶学性质的研究。该酶被纯化了1.83倍。该酶反应的最佳温度为35℃,最佳pH为6.0;在pH值为6.0时和低于40℃较稳定;70℃时,酶活性基本全部丧失。1 mmol/L的Ba^(2+)、Al^(3+)、Ca^(2+)、Mg^(2+)、Cu^(2+)和Zn^(2+)对酶活无明显的抑制作用,1 mmol/L的Ag^+使酶活力完全丧失。 β- glucosidase contents of eight wheat varieties were determined and it was found that wheat variety Dongnong 91 - 5992 possessed a high content of β - glucosidase. A study on the isolation and characterization of the β - glucosidase from wheat variety Dongnong 91 - 5992 was carried out and the isolated enzyme was purified 1.83 fold. Resuits: The enzyme exhibits optimum activity at 35 ℃ and pH 6.0; it performances normally at pH 6.0 and 〈 40 ℃, while is mostly inactivated at 70 ℃. The β - glucosidase is not obviously inhibited by Ba^2+ , Ca^2+ , Al^3+ , Mg^2+ , Cu^2+ and Zn^2+ of lmmol/L, and is completely inactivated by Ag + of 1 mmol/L.
