摘要 采用中性蛋白酶直接酶解杜仲籽仁乳液、有机试剂萃取杜仲籽油,通过正交试验优化提取工艺条件。结果表明,酶法提取杜仲籽油的水浸提工艺为:固液比1:5、pH值7.5、60℃浸提1.5 h;酶解工艺为:酶用量1 600 U/g、pH值7.5、55℃酶解2.5 h;溶剂萃油工艺为:石油醚用量1:3,萃取介质pH4.0,45℃萃取20 min;采用最佳工艺进行重复试验.提取率高达87.3%。酶法提取率高、外观性状和油脂品质好,工艺简单,成本较低,条件温和,为提取优质杜仲籽油、实现脱脂粕综合利用的首选方法。 Eucommia seed oil was obtained by directly hydrolyzing Eucommia seed milk with AS. 1398 neutral protease and then extracting oil with petroleum ether. The extraction technology condition was optimized by an orthogonal experiment. The aqueous extraction conditions are ratio of solid to liquid 1:5, soaking in water of pH 7.5 and 60 ℃ for 1. 5h. The enzymatic hydrolysis conditions are enzyme dosage 1 600 U/g, hydrolyzing in Eucommia seed milk of pH 7.5 and 55 ℃ for 2.5h. The optimum conditions of extracting oil with petroleum ether are as follows; firstly stopping enzyme hy- drolysis in pH 4.0, petroleum ether concentration 1 : 3, then extracting at seed milk of pH 4.0 and 45 ℃ for 20 min. On these optimumconditions, the oil extraction yield runs out at 87.3 %. The method of enzymaticextraction exhibits the advantages of high oil yield, better oil sensory characteristics and oil quality, simple technology, lower produce cost, and mild conditions. It is a preferred method to extract high grade Eucommia seed oil and to realize comprehensive utilization of the defatted meal.
