摘要
以盐生植物盐芥为材料,依据拟南芥中DREB1E的序列信息设计引物,扩增出盐芥中DREB1E的部分序列,然后使用SMARTTM RACE等方法,从盐芥中克隆到全长的DREB1E cDNA序列。将这一基因命名为ThDREB1E。同源性分析结果表明,ThDREB1E与拟南芥DREB1E cDNA编码区的同源性为83.78%;根据基因核酸序列推测的氨基酸序列同源性为81.18%。
Dehydration responsive element binding 1E(DREBIE),induced to express in high-salt stress,is a member of CRT/DRE binding factors (CBFs) in Arabidopsis. A partial gene homologue to DERB1E was obtained from Thellungiella halophila using PCR-based technique with a pair of primers designed on the basis of the DREBIE gene of Arabidopsis. Then,the full length of the gene was cloned from Thellungiella halophila by SMARTTM RACE based on the sequence information of the partial gene sequence. It is designated as ThDERB1E. The ThDREB1E shares 83. 78% identity in the coding se- quence of cDNA and 81.18% identity in putative amino acid sequence with that from Arabidopsis. This result will be helpful for further characterization of the expression pattern and the function of the ThDERBIE in Thellungiella halophila.
