摘要
目的:探讨快速 HPLC 分析方法在重组人胰岛素肽图测定中的应用。方法:采用 Shimadzu Pack XR—ODS(3.0 mm×75mm,2.2 μm)色谱柱;流动相 A 为0.2 mol·L^(-1)硫酸盐缓冲液(pH 2.3)-乙腈(90:10),流动相 B 为乙腈-水(50:50),进行梯度洗脱。检测波长为214 nm;柱温50℃;流速0.8 mL·min^(-1)。结果:人胰岛素酶解样品的各个片段能够很好地分离,分析时间显著缩短。结论:本法简便、快速,经济,灵敏度高。可应用于重组人胰岛素肽图的测定。
Objective:To discuss the application of fast HPLC analysis method in human insulin peptide mapping. Method:The column was Shimadzu Pack XR-ODS(3.0 mm ×75 mm,2.2 μm),0.2 mol·L^-1 sulphate buffer - acetonitrile(90: 10) and acetonitrile-water(50: 50) were used as mobile phase A and mobile phase B ,respectively. Performing the gradient elution process. The detection wavelength was 214 nm, column temperature was 50℃ and flow rate was 0. 8 mL·min^-1. Results:The digested fragments of human insulin were well separated, analysis time was shorten remarkably. Conclusion:This method is simple, economic, rapid, high sensitive and reliable for testing of human insulin peptide mapping.
