高大毛霉液体发酵培养基的优化及产酶特性研究 被引量:5

摘要 采用正交实验确定高大毛霉(MHC-7)最佳发酵培养基组成为:黄豆粉3%、可溶性淀粉0.4%、硫酸镁0.2%、磷酸二氢钾0.2%、蔗糖0.3%和黄泔水。通过对MHC-7胞内蛋白酶、α-淀粉酶、α-半乳糖苷酶、谷氨酰胺酶、纤维素酶、脂肪酶活力测定,结果表明,产蛋白酶、α-淀粉酶、α-半乳糖苷酶和谷氨酰胺酶的峰值时间分别是36、48、42、48h,纤维素酶和脂肪酶在72h内酶活力一直呈上升趋势。 Applying an orthogonal experiment to obtain the best fermentation medium comprising of Mucor mucedo (L) Fres ( MHC-7), the result were: soybean powder 3%, soluble starch 0.4%, MgSO4 0.2%, KH2 PO4 0.2%, sucrose 0.3% and wastewater of bean products. By determination of MHC-7 intracellular proteinase,α-amylase,α-galactosidase, glutaminase, cellulose, lipase, the peak time of proteinase, α-amylase, α-galactosidase, glutaminase were 36, 48,42,48h and by adopting phosphate buffer, cellulase and lipase of the activity was on the upward trend within 72h.
