广东地区汉族人HLA-DQB_1基因对胃溃疡患者的遗传易感性研究 被引量:2

摘要 目的探讨广东地区汉族人人类白细胞抗原-DQB1(HLA-DQB1)基因与胃溃疡(GU)的遗传关联。方法应用HLA基因的聚合酶链反应-序列特异性引物(PCR-SSP)核苷酸分型技术,对135例GU患者和105例正常人的HLA-DQB1等位基因进行分析。结果GU患者HLA-DQB1*0602等位基因频率与正常对照组比较明显增高,差异有显著性(P=0.001,RR=0.115,Pc=0.014),而GU患者HLA-DQB1*0604等位基因频率与正常对照组比较明显降低,差异有显著性(P=0.001,RR=17.290,Pc=0.014)。结论HLA-DQB1*0602基因对GU患者有遗传易感作用,而HLA-DQB1*0604基因对GU患者有遗传抵抗作用,GU患者与正常人之间存在着免疫遗传异质性的差异。 Objective To study the genetic susceptibility of HLA-DQB1 alleles in gastric ulcer in Chinese Hans from Guangdong and its nearby regions.Methods 135 patients with gastric ulcer and 105 healthy controls were genotyped for HLA-DQB1 by polymerase chain reactions with sequence specific primers(PCR-SSP).Results The allele frequency of HLA-DQB10602 in patients with gastric ulcer(59.3%)was significantly higher than that in healthy controls(14.3%).In contrast,the alleles frequency of HLA-DQB10604 was significantly lower in patients with gastric ulcer(0.7%)than that in healthy controls(11.4%).Conclusion These results show that HLA-DQB10602 is a susceptible gene for gastric ulcer in Guangdong Hans while HLA-DQB10604 has genetic resistance function and suggest that there are immunogenetic differences in HLA-DQB1 locus between gastric ulcer patients and healthy controls.
