摘要
目的:探讨应用来曲唑促排卵对大鼠围着床期子宫内膜整合素αvβ3和HOXA10表达的影响。方法:将60只Wistar雌鼠随机等分为3组:来曲唑组(letrozole,LE组),克罗米芬组(clomiphenecitrate,CC组),生理盐水组(NS组),分别进行促排卵。用免疫组化法观察大鼠围着床期子宫内膜腺上皮整合素αvβ3的表达变化;应用RT-PCR和Westenblot方法观察大鼠围着床期子宫内膜HOXA10表达情况。结果:LE组大鼠子宫内膜αvβ3表达与NS组无差异(P>0.05),但LE组显著高于CC组(P>0.01);NS组HOXA10表达高于LE组(P>0.05),LE组HOXA10表达高于CC组(P>0.01)。结论:来曲唑促排卵对大鼠子宫内膜容受性的抑制作用比克罗米芬小,有可能成为较为合适的促排卵药物之一。
Objective: To investigate the effects of letrozole on expression of integrin αvβ3 and HOXA10 on endometrium in rats. Methods: Sixty sexually mature female Wistar rats were randomly divided into 3 groups: LE group was given letrozole (2 mg/kg × 2 d), CC group was given clomiphene citrate(100 μg/kg × 2 d), and NS group was given 0.9% NS of the same volume. Immunohistochemical of ABC methods using monoclonal antibody against integrins αvβ3 were performed on endometriurrL Reverse transcription polymerase chain reaction (RT-PCR) method and Western blot were utilized to detect and quantitate HOXA10 mRNA expression levels in the endometrium. Results: The expression ofintegrins αvβ3 in CC group was significantly lower than those in LE group and NS group (P〈0.01). HOXA10 expression in NS group was significantly the highest among 3 groups (P〈0.05), and that in LE group was higher than that in CC group (P〈0.01). Conclusion: In rats, letrozole has less suppression on endometrial receptivity than clomiphene. In the future, letrozole may act as an appropriate drug in ovulation induction.
