优变梨橙材料的RAPD分析

摘要 永川梨橙1号和永川梨橙5号是优良芽变材料,为确定这两份材料的性状究竟属于遗传变异还是环境饰变及其来源,实验采用RAPD对永川梨橙1号、永川梨橙5号和普通梨橙、锦橙一起进行了分析.结果表明:在70个引物中有6个引物可扩增出条带,共计94条,其中多态性条带数为27条,平均每个引物出现多态带4.5条.特别是引物J039、S150和S304可清晰的区分开这4份材料.聚类分析发现,梨橙和锦橙遗传相似系数在90%左右,永川梨橙1号与普通梨橙遗传上最为相似,而永川梨橙5号与梨橙亲缘关系则相对较远,表明其发生了较大程度的变异,这与其生物学性状的表现是吻合的. Yongchuan Licheng 1 and Yongchuan Licheng 5 are high quality mutants of Licheng orange selected by Fruit Research Institute of Chongqing Academy of Agriculture Sciences in Yongchuan. To make sure whether any genetic variation occurred in them and to identify its origin, Licheng, Jincheng and the two mutants of Licheng orange were studied by random amplified polymorphic DNA (RAPD). Six of sev- enty random primers produced 94 bands, 27 of which were polymorphic, or 4. 5 polymorphic bands per primer. Several specific bands amplified by the primers J039, S150 and S304 were especially valuable for identifying the four materials studied. Cluster analysis showed that the similarity coefficient between Licheng and Jincheng orange was about 90%, and that between Yongchuan Licheng 1 and normal Licheng was the highest in all materials, but the similarity coefficient between Yongchuan Licheng 5 and normal Li- cheng was the lowest, which indicated a high level variation. The result of cluster analysis was consistant with that of morphological characters.
