摘要 目的研究不同浓度的纯化重组人 C 反应蛋白(rhCRP)对正常人血小板内皮型一氧化氮合酶(eNOS)活性的影响。方法取健康人外周静脉血,用凝胶色谱柱法收集血小板,用同位素二步色谱法测定血小板eNOS 的活性。将收集的血小板和组织胺、N_ω硝基左旋精氨酸甲酯盐酸盐(L-NAME)、不同浓度纯化的 rhCRP-起孵育后测定血小板 eNOS 活性。结果组织胺可以明显增加血小板 eNOS 活性;eNOS 抑制剂 L-NAME 可以明显抑制 eNOS 活性以及用组织胺刺激后的血小板 eNOS 活性增加;rhCRP 明显抑制组织胺刺激后的 eNOS 活性,且这种抑制作用呈现显著的浓度依赖性。结论 rhCRP 对正常人血小板经组织胺诱导的 eNOS 活性有显著抑制作用,这种抑制作用一定范围内呈现显著的浓度依赖性。 Objective To study the effect of recombinant human C-reactive protein (rhCRP) on platelet endothelial nitric oxide synthase (eNOS) activity. Methods Peripheral venous blood was collected and platelets were isolated with gel-filtration chromatography and incubated with histamine. L-NAME and different concentrations of rhCRP(5, 10, 25, 50, 100 mg/L) were co-cultured with platelet for 30 minutes, eNOS activity was measured by production of 3 H-L-citrulline from 3 H-L-arginine. Results Platelet eNOS activity was significantly inhibited after incubated platelets with L-NAME and increased after incubated with histamine (P〈0. 05). rhCRP significantly inhibited eNOS activity increase by histamine in a concentration-dependent manner (P〈 0.05 ). Conclusion rhCRP significantly inhibits eNOS activity induced by histamine in platelets in a concentration-dependent manner.
