中药对模拟失重大鼠骨骼-肌肉系统干预的初步研究 被引量:12

Preliminary Study on Effect of Chinese Herb Medicine Compound on Bone-muscle System in Rats under Simulated Weightlessness

摘要 目的研究中药复方对3周模拟失重大鼠一般状况及骨骼-肌肉系统改变的干预作用,初步观察在模拟失重情况下,复方对钙剂(牡蛎醋酸水解物)的协同作用。方法雄性Wistar大鼠30只,随机分为空白组、模型组、中药组,3组皆于悬吊笼单独饲养,空白组始终自由活动,模型组、中药组适应饲养1周后,尾部悬吊3周模拟失重。中药组每日灌胃给予中药复方水煎剂,空白组、模型组每日给予等量牡蛎醋酸水解物水溶液。观察中药复方对模拟失重大鼠体重、腓肠肌和比目鱼肌重量和指数,以及股骨重量、长度、密度、力学强度、无机质及有机质含量的影响。结果在3周模拟失重的中后期,模型组体重显著下降(P<0.01);模拟失重3周后,后肢肌肉重量与指数显著下降(P<0.01),股骨湿重、干重、有机质重、无机质重、骨密度、力学强度均显著降低(P<0.01);中药组体重较模型组显著提高(P<0.01),腓肠肌、比目鱼肌重量显著提高(P<0.01或0.05),骨密度及成分含量显著提高(P<0.01或0.05),骨力学强度有改善趋势。结论中药复方可显著改善3周模拟失重所造成的大鼠一般状况和骨骼-肌肉系统改变;单纯补充钙剂不足以对抗模拟失重所致骨丢失,中药其它药味可能对钙剂牡蛎起到了协同作用。 Objective To study the effect of Chinese herb medicine compound on general state and bone-muscle system in simulated weightlessness rats, and to observe the synergistic action of other ingredients in the compound on calcium. Methods Thirty Wistar rats were divided into 3 groups: control group, tail suspend group, taft suspend and medicine group which were given Chinese herb medicine compound by intragastric administration. After 3 weeks simulated weightlessness, body weight (BW), muscle weight (MW) and index (MI) of posterior limb, bone length (BL), wet weight (BWW), index (BI), dry weight (BDW), content of organic (ORG) and inorganic (INO) substance, bone mineral density (BMD), and mechanical properties (MEC) of femur were observed. Results At the middle-later stage of the experiment, BW of taft suspend group decreased significantly (P〈0.01). After 3 weeks simulated weightlessness, MW and MI, BWW, BI, BDW, ORG, INO, BMD, and MEC of femur all decreased significantly (P〈0.01) in tail suspend group compared with control group. In rats of tail suspend and medicine group, BW increased significantly (P〈0.01), MW and MI also increased significantly (P〈0.01 or 0.05), bone properties like BWW, BDW, ORG, INO, BMD increased significantly (P〈0.01 or 0.05), and MEC tend to increase compared with tail suspend group. Conclusion Chinese herb medicine compound is effective to prevent the change of muscle, bone and general body state. Simple calcium supplement can not prevent simulated weightlessness induced bone loss, and other ingredients in the compound may perform synergistic action to calcium (Concha Ostreae prepared by acetic acid).
