摘要
以蚬壳花椒组培苗的叶片和茎段为外植体,研究不同的基本培养基、不同种类和浓度的激素及不同的培养条件对蚬壳花椒愈伤组织诱导及其生长的影响.结果表明,MS为最适培养基;单独使用6-BA不能诱导出愈伤组织,但配合生长素使用有利于愈伤组织的形成.诱导蚬壳花椒愈伤组织的较优激素组合为6-BA0.5mg/L+NAA1.5mg/L+2,4-D0.1mg/L,诱导率达65.72%.在蚬壳花椒愈伤组织诱导过程中,2,4-D是最重要的因素.在培养过程中,以暗培养14d为最适暗处理时间.
In this paper, the leaves and the stems of Zanthoxyoum dissitum Hemsl were used as explant, which were added with different concentrations of 6-BA, NAA, 2, 4-D, in order to study the effect of different factors on callus induction and growth of Zanthoxyoum dissitum Hemsl. The experiment results showed that MS was the best medium, while 6-BA couldn't induce callus itself. And the callus could grow quickly with proper NAA and 2, 4-D. The best medium for high efficient callus induction was 6-B A 0.5 mg/L + NAA 1.5 mg/L + 2,4-D 0.1 mg/L and the frequency was 65%. In the cause of inducting callus of Zanthoxyoum dissitum Hemsl, 2, 4-D was the most important factor. Among them, the optimum condition was cultivated in darkness in 14 days.
