摘要
目的探讨Apoptin质粒对人膀胱癌裸鼠皮下移植瘤的抑瘤作用。方法皮下注射T24细胞,建立裸鼠移植瘤模型,于肿瘤直径0.5cm大小时,多点注射脂质体包裹质粒pApoptin-EGFP,设立空质粒组、生理盐水组作为对照组,观察裸鼠肿瘤生长情况;5周后处死,绘制肿瘤生长曲线,计算抑瘤率,肿瘤组织石蜡包埋,常规切片,TUNEL法检测移植瘤凋亡情况。结果成功建立移植瘤模型,注射脂质体包裹pApoptin-EGFP质粒能明显抑制移植瘤生长;TUNEL检测凋亡率为(23.24±6.12)%,与对照组相比,差异有统计学意义(P<0.01)。结论Apoptin通过诱导凋亡能明显抑制人膀胱癌裸鼠皮下移植瘤生长。
Objective To investigate the antitumor efficacy of intratumoral administration of plasmid expressing Apoptin in athymie mouse T24 bladder tumor exnograft models grafted subcutaneously. Methods Established athymie mouse human bladder tumor transplantation tumor modelby injeeting T24 eells subeutaneously; To observe the growth of athymie mouse transplantation tumor,when tumor diameter was about 0.5cm,Apoptin-EGFP plasmid pareled with liposome injeeted in it, empty plasmid group and physiologieal saline group were established as control groups,The tumor weight was measured and caleulated eontinually, The miee were killed five weeks later, tumor suppression rate was evaluated and the apoptosis index of eells was deteeted by in situ TUNEL. Results Established athymie mouse exnograft tumor model sueeessfully, the Apoptin-EGFP group eould inhibit the growth of tumor obviously eontrasted to the eontrols,the apoptosis rate was (23.24±6.12)%. Conclusion Apoptin eould inhibit obviously the growth of athymie mouse human T24 bladder eareinoma transplantation tumor by indueing apoptosis.
