摘要
目的:通过基因芯片对人血样本的抗高血压药物相关基因进行检测分型。方法:快速提取人血样本脱氧核糖核酸(DNA),对5个目标位点(CYP2D6、CYP2C9、ADRB1、AGTR1和ACE)进行特异序列扩增,扩增产物与基因芯片杂交后扫描,判断基因型。结果:志愿者的5个高血压相关基因位点分别是WW、WM、WW、WW和Ⅱ型。结论:该方法简便快捷,稳定性好,可以作为临床高血压个体化用药的遗传信息参考。
OBJECTIVE: To detect and genotyping the antihypertensive drug related genes in human blood samples by using gene chip. METHODS: DNA was extracted rapidly from human blood samples and specific amplification of five target sites (CYP2D6,CYP2C9,ADRB1,AGTR1 and ACE) was performed. Then the amplification product was hybridized with gene chip for genotyping by scanning. RESULTS: The five antihypertensive drug related gene locuses were WW, WM, WW, WW and Ⅱ . CONCLUSION : This method is simple, rapid and stable, and it serves as a genetic reference for clinical individualized antihypertensive drug therapy.
