摘要
目的建立用亲和层析处理的抗体鉴定布鲁氏菌致病株与疫苗株差异抗原的方法。方法以羰基二咪唑(CDI)活化载体sepharose CL4B,制备布鲁氏菌抗原亲和层析柱。致病株M28抗体先经疫苗株M5抗原柱吸附、中和,洗出液再经致病株M28抗原柱纯化、浓缩,分别与M28、M5抗原进行免疫印迹反应。结果处理后的M28抗体与M28抗原间存在两条特异的反应条带,此两条条带没有出现在与M5抗原的反应条带中。结论通过亲和层析处理的致病株M28抗体能够用于布鲁氏菌致病株与疫苗株差异抗原的鉴定。
Objective Establish a method to identify the different antigens between the wild-type Blucella strain and the vaccine strain by antibody treated with affinity chromatography. Methods Use carbonyl diimidazole (CDI)-activated Sepharose CL-4B to prepare affinity column. Add the antibody of wild-type Blucella strain M28 to the Ms antigens column, let them flow through the column, then add them to the M28 antigens column, collect the antibody, do the immunoblotting assay with M28 antigens and M5 antigens respectively. Results Two specific bands showed in the immunoblotting assay between the M28 antibody and M28 antigens, and the bands were inexistence in the immunoblotting assay between the M28 antibody and M5 antigens. Conclusion The antibody of M28 treated with affinity chromatography could be used for identifying the different antigens between the wild-type Blucella strain and the vaccine strain.
