摘要
目的建立大鼠慢性高眼压模型,观察不同时期的眼压变化、视神经的损伤情况和玻璃体中游离谷氨酸浓度的变化。方法Wistar大鼠30只,分为3组。用波长为532nm的氪离子黄绿激光光凝大鼠小粱网,光凝前测量一次眼压,以后每周测量眼压。在第3、6、9周分别处死1、2、3组大鼠做全视网膜铺片,1%甲苯氨蓝染色,记数平均视网膜神经元细胞密度;取20μL玻璃体用高效液相色谱分析仪测量游离谷氨酸盐浓度。结果对照眼平均眼压为(13.25±4.10)mmHg,光凝眼第1~9周平均眼压升高了1.73~2.17倍。对照眼视网膜神经元密度值为2516±196,第3、6、9周光凝眼分别为2124±108,1865±136,1654±125,差异均有统计学意义(P=0.037);第3、6、9周光凝眼玻璃体游离谷氨酸盐浓度分别为(20.10±2.45)μmol/L,(22.35±2.75)μmol/L,(23.56±2.80)μmol/L,对照眼为(18.32±2.3)μmol/L,差异均无统计学意义(P=0.661)。结论激光光凝大鼠小梁网后,眼压中等程度升高;大鼠慢性高眼压模型的视神经损害表现为视网膜神经元密度值逐渐下降,玻璃体中游离谷氨酸盐浓度变化不明显。
Objective Glutamate is thought to be responsible for the death of retinal ganglion cells that are characteristics of glaucoma. The purpose of this paper was to investigate the damage of retinal optic nerve in chronic ocular hypertension rat and the changes of free glutamate concentration in vitreous humor. Methods Experimental glaucoma was induced unilaterally in the fight eyes of 30 health adult Wister rats using a Krypton laser with wavelength of 532 nm aimed at the trabecular meshwork,and the left eyes were as control. Intraocular pressure(IOP) was measured using the Tono-Pen before and immediately after surgery and then at one week interval. 1% toluidine blue staining of whole mounted retina was utilized to label retinal nerve cells ,and these cells were counted in the 3rd, 6th and 9th week respectively. Rat models were sacrificed in the 1 st, 2nd and 3rd group rats. The free glutamate concentration in vitreous humor was measured by High-Performance Liquid Chromatographic Analysis(HPLCA). Results IOP of the experimental eyes was higher than that of the control eyes. The density value of the retinal ganglion cells in the experimental eyes was 2 124 ±108/mm^2,1 865± 136/mm^2 and 1 654 ± 125/mm^2 at the 3rd, 6th and 9th week respectively, and the value of control eyes was 2 516 ± 196/mm^2, exhibiting a significant difference among the experimental eyes and the control eyes( F = 5. 496,P = 0. 037). The free glutamate concentration in vitreous humor was 20. 10 ±2.45 μmol/L,22. 35± 2. 75 μmol/L and 23.56 ± 2. 80 μmol/L at the 3rd, 6th and 9th week in the experimental eyes,and 18. 32 ± 2. 3 μmol/L in the control eyes without significant difference among the glaucoma eyes and the control eyes( F = 0. 796, P = 0. 661 ). Conclusion lOP presents a chronical and moderate increase in an injury eye of trabecular meshwork by laser. The density of retinal nerve cells appear to be a decent tendency and the free glutamate concentration in vitreous humor is gentally increased in this experimental glaucoma model.
