摘要
目的通过检测增生型糖尿病视网膜病变(PDR)、增生性玻璃体视网膜病变(PVR)和急性视网膜坏死(ARN)患者玻璃体中MMP-9的质量浓度,探讨该分子在病变中的作用。方法采用酶联免疫吸附法检测13例PDR患者,15例PVR患者和5例ARN患者玻璃体中MMP-9的质量浓度,以单纯孔源性视网膜脱离(RD)患者作为对照。结果玻璃体中MMP-9的质量浓度PDR组为(2.10±0.51)ng/mL,PVR组为(1.61±0.22)ng/mL,ARN组为(4.00±0.49)ng/mL,与RD组(1.37±0.11)ng/mL相比,差异均有统计学意义(P<0.01)。结论三种玻璃体视网膜疾病患者玻璃体中MMP-9的质量浓度都显著升高,提示MMP-9可能在病变中起一定的作用。
Objective The function of matrix metalloproteinases (MMPs) is to degrade various components of the extracellular matrix ( ECM ) and play an important role in angiogenesis. This study was to investigate the concentration of matrix metalloproteinases-9(MMP-9) in three kinds of vitreoretinopathies and explored its role in pathogenesis of these diseases. Methods Vitrectomy was performed in 13 patients with the V stage of proliferative diabetic retinopathy (PDR) , 15 patients with proliferative vitreoretinopathy (PVR) and 5 patients with acute retinal necrosis ( ARN ) , and 19 patients with rhegmatogenous detachment of retina (RD) without PVR were used as control. The concentrations of MMP-9 in vitreous samples from surgery were detected using Enzyme linked immunosorbent assay (ELISA). Results Concentration of MMP-9 in vitreous was (2. 10 ± 0.51 ) ng/mL in PDR patients, ( 1.61 ±0. 22 ) ng/mL in PVR patients and (4.00±0.49 ) ng/mL in ARN patients, showing a significant increase in comparison with that in RD patients ( 1.37 ± 0. 11 ) ng/mL(P 〈 0.01 ). Conclusion Level of MMP-9 in vitreous elevated in three kinds of vitreoretinopathies mentioned above, suggesting that MMPs play an important role in pathogenesis of these diseases.
