过氧化物酶体增殖物激活受体γC161→T基因多态性与2型糖尿病及动脉粥样硬化的相关性 被引量:1

摘要 目的:探讨武汉地区汉族人2型糖尿病及动脉粥样硬化(AS)与过氧化物酶体增殖物激活受体γ(PPARγ)C161→T基因多态性的关系。方法:应用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)检测172例武汉汉族2型糖尿病患者(根据动脉内膜厚度超声波检测分为AS组71例,非AS组101例)及138例正常健康人PPARγC161→T基因多态性,并测定相关生化指标,综合分析基因频率、等位基因频率分布及不同基因型与临床资料、生化指标的关系。结果:①糖尿病组与正常人组C、T等位基因频率分布无显著差异(P>0.05),在2型糖尿病AS组和非AS组C、T等位基因频率的分布亦无显著差异。②2型糖尿病组中CC基因型患者的甘油三酯明显高于T等位基因携带者(P<0.05)。结论:PPARγC161→T基因多态性与武汉地区汉族人2型糖尿病的发生及其AS的发生无明显相关,但T等位基因携带者与低甘油三酯相关联可能对动脉粥样硬化的发生具有保护作用。 Objective: To investigate the association of PPAR γC161→T gene polymorphism with type 2 diabetes mellitus (T2DM) and atherosclerosis(AS) in Chinese Han people in Wuhan. Methods:A total of 172 T2DM patients and 138 healthy Han people of Wuhan were investigated in this study. According to arterial intima-media thickness (IMT) measured by color Doppler ultra- sonography, all the 172 T2DM cases were divided into two groups as AS group(n=71)and non-AS group(n=101). PPAR γC161→T gene polymorphism was determined by polymerase chain reaction and restriction fragment length polymorphism, and some biochemical indicators were tested. The frequencies of PPAR γC161→T genotypes and the allele were compared as well as the clinical data. Results: There was no significant difference between T2DM group and control group, and between the AS group and non-AS group in the frequency of C and T allele. The triglyceride level in CC genotypes of the T2DM was significantly higher than that in T allele carriers. Conclusion: PPAR γC161→T gene polymorphism is not associated with the incidence of T2DM and AS in Han people of Wuhan Chinese. But T allele is associated with lower level of tri-glyceride, and could influence the process of atherosclerosis.
