大鼠输注经紫外线照射的供体脾细胞后神经移植中T淋巴细胞亚群的变化 被引量:1

摘要 目的:通过尾静脉预输注经紫外线照射的供体脾细胞后进行同种异体坐骨神经移植,观察免疫排斥过程中大鼠T淋巴细胞亚群的变化,探讨周围神经缺损修复的可行性。方法:实验分自体神经移植组(自体移植组、A组)、异体神经移植组(异体移植组、B组)、尾静脉预输注经紫外线照射的供体脾细胞后进行异体神经移植组(后行移植组、C组)。分别在移植术后第3d、第7d、第14d、第28d,用荧光标记CD4、CD8单抗作用后,流式细胞仪检测T淋巴细胞及其亚群百分数和CD4/CD8比值。结果:与A组比较,第7d,B组和C组小鼠外周血CD8阳性细胞百分率和CD4/CD8比值显著增高(P<0.05,P<0.01);第14d,B组小鼠外周血CD4、CD8阳性细胞百分率和CD4/CD8比值明显增高(P<0.05,P<0.01),C组小鼠外周血CD4、CD8阳性细胞百分率明显增高,CD4/CD8比值明显下降(P<0.05);第28d,B组和C组小鼠外周血CD4、CD8阳性细胞百分率和CD4/CD8比值明显增高(P<0.05,P<0.01)。结论:紫外线照射供体特异性抗原可以有效地诱发机体产生免疫耐受,从而减轻免疫排斥反应。 Objective:To observe the changes of subgroups of T lymphocyte in peripheral plasma of rats after nerve transplantation.Methods:96 rats were randomly divided into group A,group B and group C.Group A and group B progressed nerve autograft and nerve allograft respectively. Suspension of splenocytes of donor rats were irradiated by UVB, then the rats in group C were injected with irradiated suspension of splenocytes through caudal vein, then the transplantation of the sciatic nerve from donor rats to the rats in group C was done 1 week later.After nerve transplantation, the percentages of CD4 and CD8 T lymphocytes and the ratio of CD4 to CD8 in peripheral plasma of the recipient rats were detected by flow cytometry.Results:Compared with group A,on the seventh clay,the percentage of CD8 T lymphocytes and the ratio of CD4 to CDs in group B and C were increased significantly;on the fourteenth clay,the percentages of CD4 and CD8 T lymphocytes and the ratio of CD4 to CD8 in group B were increased significantly,the percentages of CD4 and CDs T lymphocytes in group C were increased,hut the ratio of CD4 to CD8 in group C was decreased;on the twenty-eighth day, the percentages of CD4 and CD8 T lymphocytes and the ratio of CD4 to CD8 in group B and C were increased significantly. Conclusion :Suspension of splenocytes of donor rats were irradiated by UVB could relieve the immunogenicity of allograft and alleviate immunological rejection of recipient rats.
