膀胱肿瘤T24/ADM细胞多药耐药模型的建立及其耐药性的鉴定 被引量:4

摘要 目的建立人膀胱肿瘤T24/ADM细胞多药耐药系以及对其耐药性进行鉴定。方法用阿霉素(ADM)浓度梯度递增诱导法培养膀胱肿瘤T24/ADM多药耐药细胞。用四甲基偶氮唑蓝(MTT)法测不同药物对膀胱肿瘤T24/ADM细胞多药耐药性,流式细胞术定量检测细胞P糖蛋白(P-gp)的表达变化。结果T24/ADM细胞对ADM有明显的耐药性,耐药指数(RI)16.3,并对其他药物也产生耐药性。T24/ADM细胞P-gp的表达率明显增高(P<0.01)。结论经鉴定人膀胱肿瘤T24/ADM细胞具有多药耐药特性。 Objective To establish multidrug resistance model in bladder tumor T24/ADM cell lines and to assess their resistant characteristics, Methods Human bladder tumor cell line T24 was induced to multidrug resistance cell line T24/ADM by intermittent administration of high dose (ADM Adriamycin), The multidrug resistance characteristics was evaluated by MTT assay; P-gp expression was determined by flow cytometry, Results T24/ADM has drug resistance characteristics to ADM, and its IC50 of ADM was 16.3 times higher than that of T24. The expression of P- gp in T24/ADM was markedly higher ( P 〈 0.01 ), Conclusion Multidrug resistance bladder tumor T24/ADM cell lines has been established and T24/ADM is a human multidrug-resistant cell line.
