摘要
目的检测谷胱甘肽巯基转移酶(GSTP)1基因多态性与中国人前列腺癌相关性。方法收集重庆地区前列腺癌血标本81例,50岁以上对照组血标本90例,从外周血提取DNA,采用聚合酶链反应(PCR)分析GSTP1基因型分布频率。结果GSTP1105Ile/Ile、Ile/Val、Val/Val三种基因型分布频率分别为51%、38%和11%,与对照组64%、32%、3%相比,105Val/Val基因型分布频率显著增高(P<0.05,OR=4.244,95%CI=1.082~16.642),3种基因型均与前列腺癌分期、分级无关。结论GSTP1105Val/Val基因型与中国人前列腺显著相关,GSTP1基因多态性与前列腺癌分期、分级无关。
Objective To observe the relationship between the glutathione S-transferase genes pi (GSTP1) and the risk of prostate cancer (PCa) in Chinese population. Methods We collected blood samples from 81 prostate cancer patients and 90 controls aged above 50 in Chongqing. DNA was extracted from each blood sample, the frequencies of GSTP1 genotypes were analyzed using polymerase chain reaction, restriction fragment length polymorphism (PCR- RFLP), Results The frequencies of the three gene type GSTP1 105 Ile/Ile, Ile/Val, Val/Val were 51%, 38 % and 11% respectively in patients of prostate caner and 64% ,32% and 3% in controls. It showed that the prevalence of Val/Val gene type in PCa was significantly higher than that in controls (P 〈 0.05, OR = 4.244,95% CI = 1.082 -16.642), None of the three gene types was found to beassociate with disease status and Gleason grade of PCa. Conclusion GSTP1-105Val/Val gene type was found to be significantly associate with the risk of prostate cancer in Chinese population. No statistically significant associations were found between polyrnorphism of GSTP1-1105V with disease status and Gleason grade of PCa.
