心可舒片对血管内皮细胞凋亡及凋亡调控基因的影响 被引量:4

摘要 目的:探讨血管紧张素Ⅱ(AngⅡ)在不同浓度和不同作用时间下对内皮细胞凋亡率及凋亡调控基因的作用及心可舒片对其影响。方法:采用流式细胞技术测定了在血管紧张素Ⅱ不同浓度和不同作用时间下内皮细胞凋亡率及凋亡基因Fas和Bcl-2表达量的变化和应用药物后对其的影响。结果:血管紧张素Ⅱ可明显促进凋亡率及凋亡基因的表达量,且其作用呈剂量依赖性和时间依赖性;经药物处理后的内皮细胞对AngⅡ的反应完全不同,凋亡率明显降低,同时Fas的表达量随药物浓度增加和作用时间延长而降低,Bcl-2则增高。结论:血管紧张素II具有明显的促进细胞凋亡和凋亡基因表达的作用;心可舒片对细胞凋亡和凋亡基因表达有一定的调节作用。 Objective : To study the effects of angiotensin Ⅱ (AnglI) in different concentrations and different action time on the ratio of apoptosis and expression of apoptosis genes in vascular endothelial cells and the influences of Xin-keshu tabelet on it. Methods : Flow cytometer was used to measure the apoptosis rate and the expression of apoptosis genes (Fas and Bcl- 2 ) induced by AnglI in different concentrations and different action time, and influenced by the drug. Results : The apoptosis rate and the expression of apoptosis genes were induced and increased by AnglI with the increase of concentrations and action time compared with the control. The expression of Fas was decreased and Bcl - 2, increased with the increase of the concentrations and action time of AnglI after the uses of drugs compared with AnglI group. Conclusion: The apoptosis rate and the expression of apoptosis genes may be induced and increased by angiotensin Ⅱ with the increase of the concentrations and the action time. The use of drug may have the regulating effect on the apoptosis rate and the expression of apoptosis genes in vascular endothelial cells.
