益肾活血汤对糖尿病大鼠肾功能保护作用的实验研究 被引量:1

摘要 目的:探讨益肾活血汤对糖尿病大鼠肾功能的保护作用及其机制。方法:将雄性SD大鼠分为4组:正常对照组(A组)、低剂量组(B组)、高剂量组(C组)、糖尿病组(D组),在实验的第3周、第6周分别测定尿中微量白蛋白(mAlb)含量和β-N-乙酰氨基葡萄糖苷酶(NAG)活力和血清中血糖(GLU)、甘油三酯(TG)、血肌酐(SCr)、尿素氮(BUN)、糖化蛋白(GSP)丙二醛(MDA)含量及超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活力。结果:糖尿病组大鼠首先出现尿中mAlb含量增高,随病程的进展NAG活力也开始升高。益肾活血汤可明显降低尿中mAlb含量,对NAG活力的影响不大。此方剂无明显的降血糖作用,但可明显降低TG和BUN的含量。从第3周开始,糖尿病大鼠的GSP、MDA含量明显高于正常组,而SOD、GSH-Px活力明显低于正常组,到第6周,益肾活血汤能够降低GSP、MDA含量,提高SOD、GSH-Px活力。结论:糖尿病大鼠在肾功能完好时,已开始出现mAlb增高,而后才出现NAG活力增高,糖尿病大鼠体内存在脂代谢紊乱、非酶糖化反应、氧化应激增强,这些是导致糖尿病肾病的原因,益肾活血汤能够通过抗脂代谢紊乱、非酶糖化反应、氧化应激增强来保护肾功能。 Objective: To assess the benefit kidney and activate blood Chinese herb effects of the renal function in kidneys of the diabetic rats and mechanism. Methods: SD rats were induced by streptozocin(STZ) to make diabetic model rats ,the diabetic rats were randomly divided into four groups:normal control group(A) ,low dose group( B ,benefit kidney and activate blood Chinese herb 18g · kg^-1 · d^-1 by gavage) ,high dose group(C,26g· kg^-1· d^-1 by gavage) ,diabetic model group (D). At week 3,6 respectively the contents of mAlb and activities of NAG in urine and the contents of blood glucose, triglyeerides (TG), serum ereatinine (SCr), blood urea nitrogen ( BUN ), glyeasylation products ( GSP), lipid peroxidatin(MDA) in serum and the activities of superoxide dismutase(SOD) ,glutathione peroxidase( GSH -P, ) in serum were determined. Results :Before damaged of renal function in kidneys of diabetic rats ,the contents of mAlb began to increase. Benefit kidney and activate blood Chinese herb could reduce the contents of mAlb, but could not change the activities of NAG and the contents of blood glucose. Benefit kidney and activate blood Chinese herb could reduce the contents of TG from 3 weeks. The contents of GSP and MDA of diabetic rats were higher than the contents of GSP and MDA of normal rats. the activities of SOD, GSH - P, of diabetic rats were lower than the activities of SOD, GSH - P, of normal rats. Contusion: The change of the renal function in kidneys of the diabetic rats begin from the contents of mAlb in urine, lipid metabolism disorder, nonenzymatie glyeation,the increased oxidative stress in diabetic rats are the reasons of result in diabetic nephropathy(DN). Benefit kidney and activate blood Chinese herb can protect the renal function in kidneys of the diabetic rats by reducing nonenzymatie glycation and oxidative stress and the contents of mAbl and improving lipid metabolism.
