蒺藜皂苷对PC12细胞氧化应激损伤的保护作用 被引量:5

摘要 目的探讨蒺藜皂苷(GSTT)对H2O2诱导的PC12细胞氧化应激损伤的保护作用。方法用H2O2刺激PC12细胞使其发生氧化应激损伤,MTT法检测PC12细胞活性,流式细胞仪检测PC12细胞凋亡率及线粒体膜电位变化,Hoechst33258染色观察PC12细胞核形态,琼脂糖凝胶电泳观察DNA ladder图谱。结果与模型组比较,GSTT各剂量组可使PC12细胞存活率提高(P<0.001,P<0.01),凋亡率降低,线粒体膜电位回升,细胞核浓染致密数减少,无典型DNA ladder条带,且在一定范围呈剂量依赖性。结论蒺藜皂苷对H2O2诱导的PC12细胞氧化应激损伤具有保护作用,其作用机制可能与稳定线粒体膜电位有关。 Objective To investigate the protective effect of gross saponins of tribulus terrestris(GSTT) on H2O2-induced oxidative-stress damage in PC12 cells.Methods Oxidative-stress damage of PC12 cells was induced by H2O2 at the concentration of 200μmol/L.The cell activity was analyzed by MTT assay.The cell apoptosis rate and mitochondrial membrane potential were determined by flow cytometry.The morphology changes were observed through microscope with Hoechst33258 stain.DNA ladder was observed through agarose gel electro phoresis.Results Compared with those of the model group,the cell activities of 3 GSST-treatad groups increased(P〈0.001,P〈0.01),the apoptosis rate of those three groups decreased and the mitochondrial membrane potential reinforced,typical DNA ladder could not be observed.The results mentioned above were in a dose-dependent manner.Conclusion GSTT has a protective capability to antagonize the oxidative-stress damage of PC12 cells induced by H2O2 and its mechanisms may be related to stability of mitochondrial membrane potential.
