不同浓度的黄芪总黄酮对高糖培养下牛视网膜血管周细胞凋亡的影响 被引量:8

摘要 目的探讨黄芪总黄酮对高糖培养下的牛视网膜微血管周细胞凋亡的影响。方法以在体外培养3代近融合的牛视网膜微血管周细胞为对象,分为正常对照组、高糖组(25 mmol/L)、不同浓度黄芪总黄酮1、2、3、4组(0.25mg/mL、0.5mg/mL、1.0mg/mL2、.0 mg/mL),孵育6天,采用TUNEL法检测培养后周细胞的凋亡率。结果黄芪总黄酮(0.5mg/mL、1.0mg/mL、2.0mg/mL)组周细胞凋亡率低于高糖组和对照组(P<0.01);黄芪总黄酮浓度与周细胞凋亡有关(r=0.945,P<0.01)。结论一定浓度黄芪总黄酮对高糖培养下的牛视网膜微血管周细胞凋亡有明显的抑制作用,呈剂量依赖性。 Objective To study the effect of total flavonoid of Astragalus on the apoptosis of bovine retinal capillary pericytes(BRP) under high glucose environment.Methods BRPs were cultured for 6 days respectively under control group,high glucose group(25mmol/L) and different total flavonoid of Astragalus groups(0.25mg/mL,0.5mg/mL,1.0mg/mL,2.0mg/mL).Apoptosis of BRPs were studied by in situ TUNEL method.Results The apoptosis of BRPs markedly reduced in the total flavonoid of Astragalus group(0.5mg/mL,1.0mg/mL,2.0mg/mL) compared with control group and high glucose group(P〈0.01).The change was correlated positively with the concentrations of total flavonoid of Astragalus(r=0.945,P〈0.01).Conclusion Total flavonoid of Astragalus can decrease the apoptosis of BRPs in a dose-dependent manner.
