摘要
目的探讨通过改进实验方法以提高制作心肌梗死模型的成功率。方法80只Wistar大鼠,结扎左冠状动脉前降支造成心肌梗死,分析麻醉、气管插管及呼吸道护理、开胸体位及方式、结扎位置等操作过程中的细节对大鼠存活率的影响。结果麻醉采用腹腔注射水合氯醛(300mg/kg)、气管插管应用16G静脉留置针经口直视插管、开胸方式采用右侧卧位、于左心耳和肺动脉圆锥连线下方进针结扎,通过上述方法的改进,大鼠心肌梗死模型制作成功率得到很大提高。结论随着对造模过程中相关问题的重视,提高了大鼠心肌梗死模型制作成功率,为今后的实验研究打下了基础。
Objective To elevate the survival rate of rat myocardial infarction model through ameliorated experimental conditions.Methods Left anterior descending coronary arteries were ligated to induce myocardial infarction model in 80 Wistar rats and some details that may influence the survival rate of rat were regulated.The details included the mode of anaesthesia,trachea cannula,the operation and the location of ligating to the left anterior descending coronary.Results Some methods were demonstrated to improve the survival rate of rats remarkably.These methods included intraperitoneal injection of Choral hydrate(300mg/kg),trachea cannula with 16G vein needle,opening the breast at the right recumbent position,ligating the left anterior descending coronary artery under the attachment line between the left auricle and pulmonary conus.Conclusion The survival rate of rat myocardial infarction model can be apparently elevated by ameliorating some related experimental conditions.It could provide experimental basis for further research.
