摘要
【目的】研究大鼠闭合性颅脑损伤后24h海马中蛋白质表达的变化及其特点。【方法】选成年雄性sD大鼠随机分为手术对照组及损伤后24h组,复制Marmarou落体打击闭合性颅脑损伤大鼠模型,应用双向电泳技术分析大鼠脑损伤后24h海马中蛋白质表达的改变。【结果】双向电泳分离结果显示,采用pH3~10胶条,与手术对照组比较,损伤后24h有7个蛋白质点有表达量上的差异,并有2个蛋白质点消失,新出现1个蛋白质点;采用DH4~7胶条,与手术对照组比较,损伤后24h有7个蛋白质点有表达量上的差异,并有1个蛋白质点消失,新出现3个蛋白质点。【结论】大鼠闭合性颅脑损伤早期(24h)可引起海马蛋白质表达发生变化。
[Objective] To investigate the alteration of hippocampus protein expression of rats after closed brain injury. [Methods] Male Sprague-Dawley rats were randomly divided into sham operation group and 24 h after injury group. The head injury animal model was produced by an weight-drop device designed by Marmarou, and the change of protein expression in hippocampus was detected by two-dimensional electrophoresis. [Results] In pH3 - 10 stnp, in 24 h post- injury group, there were 7 protein spots showed significant difference as compared with sham operation group. Meanwhile, 2 protein spots disappeard and 1 protein spots emerged. In pH 4 ~ 7 strip, a total of 7 protein spots 24 h post - injury group showed significant difference, moreover, 1 protein spot disappeared, and 3 protein spots emerged. [Conclusions] The alteration of protein expression in hippocampus could be induced by closed brain injury.
