检测糖化血清蛋白的试剂及方法改进 被引量:1

THE REAGENT FOR THE MEASUREMENT OF GLYCOSYLATED SERUM PROTEIN AND THE IMPROVEMENT ON METHOD

摘要 介绍三种可作为测定糖化血清蛋白的标准物,即果糖吗啉(DMF)、果糖哌啶(DPF)、a-D一葡萄糖五乙酸酯(a-D-GPA),并比较了它们的优缺点,其中后两种作为检测糖化血清蛋白的标准物,尚未见其它文献报道.通常显色剂氯化硝基四氮唑蓝(NBT)试液在存放过程中常常出现沉淀,作者通过IR和薄层色谱分析对该沉淀物进行了表征,同时对NBT的配制和储存方法进行了改进,克服了NBT在存放过程中出现沉淀的现象.选择一种既不与BNT反应又不与标准物反应,且能溶解还原产物甲的增溶剂,又使测试环境大大改善,增加了测试的重现性和稳定性. The 1-deoxy-1-morpholino-D-fructose(DMF),1-deoxy-piperidino-Dfructose(DPF) and a-D-glucopyranose pentaacetate(a-D-GPA) can be used as the standard materials to react with Nitro Blue Tetrazolium Chloride (NBT) in order to measure the glycosylated serum protein. Their virtues and defects ar also compared. DPF and a-D-GPA as the standard meterials have not been reported by other people except the authors. Generally NBT often precipitates when it is laid up; precipitation was characterized by IR and thin layer chromatograph. The method of compounding and storage of NBT has been improved, when the NBT is laid up, the precipitation phenomenon has been overcomed by one solubilizing agent (Q).Formazans can be dissoered by Q which doesn's react with NBT and standard materials. It improves the environment of test greatly, and enhances replication and stability.
