渗透处理三七细胞促使人参皂甙的有效释放 被引量:15

Effective Release of Ginseng Saponin in Cell Suspension Cultures of Panax Notoginseng Permeated with Dimethyl Sulfoxide

摘要 二甲基亚砜(DMSO)渗透处理三七悬浮培养细胞,使该细胞有效释放胞内皂甙。体积分数φ=0.02~0.05的DMSO溶液短期处理三七细胞(30min),胞内皂甙释放量为30~80mg/L;φ>0.1的DMSO溶液对细胞有严重伤害,胞内蛋白质大量放出。经φ=0.01的DMSO长期处理的三七细胞生长良好,且皂甙分泌量增加,培养基中可达130mg/L,约占皂甙总量的10%。 Dimethy sulfoxide (DMSO) was used to permeate suspension cells of Panax notoginseng for effective release of a useful intracellular secondary metabolite ginseng saponin. When the cells were treated with DMSO of φ =0.02 ̄0.05 for 30 min, the saponin released into the medium was increased from 30 to 80 mg/L. At a DMSO concentration over φ =0.1, the cells were seriously damaged and a large amount of intracellular protein was also secreted. After the cells were treated with φ =0.01 DMSO for 5 successive subcultures, the ginseng saponin released was enhanced to the highest level of 130 mg/L in medium, which reached about 10% of its total amount.
