甲基叔丁基醚对胎鼠神经元和神经递质影响 被引量:1

摘要 目的探讨大鼠妊娠期甲基叔丁基醚(MTBE)暴露对胎鼠脑组织神经元型一氧化氮合酶(nNOS)及神经递质谷氨酸(Glu)的影响。方法40只健康成年昆明种小鼠于妊娠第0 d开始至妊娠结束期间,用食用油稀释的不同浓度MTBE 0,25,100,400,800 mg/(kg.bw)进行灌胃染毒,每天1次,直到胎鼠出生。用免疫组化的方法(SABC)观察胎鼠脑组织中nNOS和神经递质Glu免疫阳性神经元的表达。结果当MTBE的剂量为400 mg/(kg.bw)时,胎鼠大脑皮质、海马和小脑nNOS和Glu阳性神经元细胞数较对照组减少(P<0.05),染色强度变浅,并存在明显的剂量-效应关系。结论妊娠期母鼠接触MTBE可致胎鼠脑组织nNOS及神经递质Glu的表达下降。 Objective To explore the effect of methyl tertiary- bitul ether (MTBT) on nNOS and neurotransmitter Glu of brain tissues of fetal mice during gestation. Methods 40 healthy and pregnant KM mice were distributed in five groups randomly. The MTBE exposed doses were 800,400,100 , 25 and 0 mg/(kg, bw) by garage from the 0 d to the end of gestation, once per day. The immunohisto - chemical method (SABC method) was used to check the expression of nNOS and neurotransmitter Glu in the brain tissues of the fetal mice. Results The expression of the nNOS and neurotransmitter Glu in the cerebral cortex, hippocampus and cerebellum of the fetal mice was dereased when the exposed dose was 400 mg/kg, bw and the data showed a dose - effect relationship. Conclusion MTBE can decreased the content and activities of nNOS and neurotransmitter Glu in the brain tissue of the fetal mice.
