硫辛酸对氢醌致V79细胞线粒体膜电位影响 被引量:1

摘要 目的利用荧光探针JC-1观察氢醌对V79细胞线粒体膜电位的影响及硫辛酸的保护性作用。方法在体外培养的V79细胞中分别加入一定浓度的硫辛酸和氢醌,培养24 h,采用倒置荧光显微镜和流式细胞术检测细胞线粒体膜电位水平。结果正常V79细胞线粒体膜电位为1.12:而50,100μmol/L氢醌可导致V79细胞线粒体膜电位下降,分别为0.22和0.03;50μmol/L的硫辛酸单独作用时不改变线粒体膜电位,但能明显拮抗50,100μmol/L氢醌引起的线粒体膜电位下降,分别为0.79和0.63。结论一定浓度硫辛酸可有效抑制50,100μmol/L氢醌引起的V79细胞线粒体膜电位的下降,对V79细胞的线粒体膜电位具有保护作用。 Objective To explore the changes of mitochondrial transmembrane potential(△ψm)in V79 cells caused by hydroquinone with fluorescent JC - 1 and the protection of lipoic acid. Methods V79 cells were cultured in vitro, then treated with lipoic acid and hydroquinone into different groups for twenty - four hours. The levels of mitochondrial transmembrane potential were analysed by flow cytometry. Results Normal V79 cells had a high△ψm(1.12), and JC- 1 aggregates were formed in the inner membrane of mitochondria and emitted orange - red fluorescence, 50μmol/L and 100μmol/L hydroquinone caused the decrease of △ψm, and JC- 1 aggregates were dissociated to monomer which emitted green fluorenscence (0.22 and 0.03), Treatment with lipoic acid alone didn't change the △ψm, but it could effectively protect V79 cells from hydroquinone toxicity(0.79 and 0.63), Conclusion The results show that lipoic acid can inhibit decrease of mitochondrial transmembrane potential of V79 cells damaged by hydroquinone.
