富锌灵芝发酵培养基的响应面法优化研究 被引量:6

Studies on the Medium Optimization for Zn-riching Ganoderma Lucidum by Response Surface Method

摘要 目的:优化灵芝发酵培养基,提高灵芝对无机锌元素的富集率。方法:运用Plackett-Burman设计对影响灵芝富锌率的营养因子进行筛选;在此基础上,利用响应面法(Response Surface Method,RSM)建立灵芝富锌率的数学模型。结果:通过Plackett-Burman试验获得了影响灵芝富锌率的3个关键因子:玉米粉、豆粕及KH2PO4;对已建立的灵芝富锌率的数学模型求解,当玉米粉38.0g/L、蔗糖20g/L、豆粕8.5g/L、Fe2(SO4)31.0g/L、NH4Cl2.0g/L、CaCO31.0g/L、MgSO41.0g/L、KH2PO41.2g/L时,可获得灵芝最大富锌率,预测值为27.06%。结论:利用RSM法优化了富锌灵芝发酵培养基,提高了灵芝的富锌率,该方法经济有效。 Objective: The liquid medium of Ganoderma lucidum was optimized to improve the Zn-enrichment. Method: The mutritive factors affecting the rate of Zn-enrichment were screened by Plackett-Burman design. Based on the result of Plackett-Burman design, response surface method was used to optimize the above critical factors and a mathematical model was built. Result: Corn flour, bean cake and KH2PO4 which were the significant factors affecting the rate of Zn-enrichment were determined by Plackett-Burman design. The optimum levels of the factors were calculated through regressive method and the result showed that the optimal conditions for obtaining 27.06% were corn flour 38.0 g/ L, sucrose 20g/L, bean cake 8.5g/L, Fe2 (SO4)3 1.0g/L, NHaC1 2.0g/L, CaCO3 1.0 g/L, MgSO4 1.0 g/L, KH2PO4 1.2 g/ L. Conclusion: Medium optimized by RSM for Ganoderma lucidurn has been proved to be conducive to raising Zn-enrichment rate and to be economic-efficient .
