摘要 目的探索-80℃条件长期保存血小板的关键技术。方法将12人份加终浓度为5%二甲基亚砜(DMSO)的富含血小板血浆,分为若干等份后置于实验专用-80℃低温冰箱中保存1—16个月,1—6个月每月每人份各取1份样本、8—16个月每2个月每人份各取1份样本37℃水浴复温后检测:血小板计数(P lt)、血小板平均体积(MPV)、血小板体积分布宽度(PDW)、pH、血小板CD62p表达、凝血酶激活CD62p表达、凝血酶激活CD62p再表达、磷脂酰丝氨酸(PS)表达和激活血小板诱导血浆凝固时间。结果在16个月的保存过程中P l、tMPV、PDW、pH、血小板CD62p表达、凝血酶激活CD62p表达、凝血酶激活CD62p再表达、PS表达和激活血小板诱导血浆凝固时间等,变化均无统计学意义(P>0.05)。结论血小板加终浓度为5%DMSO,在-80℃条件下长期保存质量较好且较稳定,此技术条件可用于长期储备血小板资源。 Objective To explore the feasibility of platelet long eryopreserved at - 80℃. Methods Aliquots of 12 fresh platelet-rieh plasmas (FPRPs) with 5% dimethyl sulphox/de were eryopreserved in the hypothermla refrigerator used specially for research for 16 months. The aliquots were thawed by 37 ℃ water bath for analysis per one month during one to six months storage and per two months during eight to sixteen months. The markers observed included platelet account, mean platelet volume, platelet distribute width, pH, CD62p express, CD62p express activated by thrombin, CD62p anew express activated by thrombin, phosphoryl serine express and clotting time reduced by the activated platelets. Results The aforementioned markers were stable and had no significant deference between the groups which thawed for analysis during the storage (P 〉 0.05 ). Conclusion Platelets maintain a good and stable quality at - 80 ℃ and the -80 ℃ refrigerator may cryopreserve the platelets for a long time.
