摘要
目的为研制筛选抗癌新药维泰醇建立适宜的动物模型。方法将有生物学活性的癌细胞接种于不同种小鼠,逐日观察、记录致瘤情况和死亡时间。结果L1210细胞腹腔接种于DBA-2、BDF-1、CDF-1、C57BL/6J和昆明鼠(KM)小鼠共86只,接种量(1×10^4-1×10^6)个/只;其中DBA-2、BDF-1和CDF-1小鼠的致瘤率为100.0%,C57BL/6J和昆明鼠(KM)的致瘤率为0。B16细胞腋窝皮下接种BDF-1小鼠20只,接种量〉1×10^6个/只,致瘤率为95.0%。MGC-803细胞腋窝皮下接种于BALB/C裸鼠,接种量(5×10^6)个/只,致瘤率为100.0%。HL(对照细胞)和Z-HL16C细胞腋窝皮下接种BALB/C裸鼠6只,接种量(5×10^6)个/0.2ml.部位,致瘤率为100.0%。结论用L1210、B16、MGC-803和Z-HL16C细胞成功制备了相应的荷瘤小鼠模型,为下一步体内药效研究打下了良好基础。
Objective To establish the suitable animal models for screening the new anti-tumour drug--Alternol. Methods We used tumour cells which had activity to the new drugs to inoculate the different kinds of mice, and recorded the tumorigenicity and death time daily. Results A total of 86 DBA-2 mice, BDF-1 mice and CDF-1 mice were given an injection into abdominal cavity respectively of L1210 cells by the quantity of 1 ×10^4-1×10^6 cells per mice. Their oncogene ratio was 100%. Twenty BDF-1 mice were inoculated subcutaneously at limb subaxilla of B16 cells by the quantity of 〉 1 ×10^6 cells per mice, and the oncogene ratio was 95 %. BALB/C nude mice were inoculated subcutaneously at limb subaxilla of MGC-803 cells by the quantity of 5×10^6 cells per mice, and the oncogene ratio was 100%. BALB/C nude mice were inoculated subcutaneously at limb subaxilla of Z-HL16C cells by the quantity of 5×10^6 cells/0.2ml each part per mice, and the oncogene ratio was 6/6. Conclusion These models might be the hopeful animal models for screening the new anti-tumour drugs in future study.
