食管内脏敏感性动物模型的制备

摘要 目的研究幼兔食管扩张后其行为学及食管黏膜上皮细胞间隙的改变,初步探讨食管内脏敏感动物模型制备的可能。方法新西兰白兔20只,随机分为3组。A组:食管扩张组,8只,每日接受直径0.9 cm的球囊扩张,共14 d;B组:对照组,6只,每日插入球囊导管,无球囊扩张,共14 d;C组:空白对照组,6只,无任何刺激。通过动物行为学评分来评价食管内脏感觉的改变;采用电镜观察幼兔食管下段黏膜上皮细胞细胞间隙的改变。结果幼兔食管机械扩张后,A组行为学评分较B、C组明显升高,有显著统计学意义(P<0.05);但是其细胞间隙无显著差异(P>0.05)。结论持续的食管扩张可以引起食管内脏敏感性增高。 Objective To observe the changes of animal activity scores and intercellular spaces of esophageal epithelial cells after esophageal balloon distention in rabbits, and to explore the possibility to prepare animal model of esophageal visceral sensitivity. Methods Twenty healthy New Zealand rabbits were randomly divided into three groups : group A ( n = 8 ), experimental group, inserting ERCP tubes with balloon (0.9 cm) into lower esophagus for fourteen days ; group B ( n = 6), control group inserting the same ERCP tubes with no balloon into the lower esophagus for fourteen days;group C (n = 6 ), control group with no insertion. The esophageal visceral senses were assessed by animal activity scores. Changes of intercellular spaces were meas- ured by transmission electron microscopy. Results The animal activity scores of group A were significantly higher than those of groups B and C ( P 〈 0.05 ). Compared with groups B and C, the diameters of esophageal epithelial intercellular spaces had no significant differences in group A. Conclusion Esophageal hypersensitivity may be induced by esophageal distention.
