低能量激光与集落刺激因子对人脐带血造血细胞的增殖作用 被引量:1

摘要 本研究用铜蒸气激光照射人脐带血造血细胞,观察低能量激光与集落刺激因子(CSF)对造血细胞的增殖作用。结果显示激光加CSF对造血细胞GMCFUc有协同增殖作用,与单用激光照射组、CSF刺激组及对照组相比,有非常显著性差异(P<0.01)。将经激光照射与未照射脐血造血细胞在液体培养基中培养2周,前者活细胞数较前增加55—110倍,后者增加11—27倍,再次软琼脂接种培养,仍有集落形成,生长的集落绝对数较液体培养前显著增加,而且激光照射组比未照射组更明显。提示低能量激光照射脐血细胞是体外扩增造血细胞的一种有效方法。 Proliferative effect of low energy laser and colony stimulating factor(CSF) on human umbilical cord blood hematopoietic cells was observed in vitro with copper vapour laser irradiation. The results showed that laser irradiation synergized with CSF in expansion of cord blood hematopoietic cell. The GMCFUc numbers from the group treated with irradiation plus CSF increased significantly as compared with that from other groups including laser irradiation or CSF treatment alone or control((P<0 01). Two weeks after incubation in liquid medium the numbers of live cell from the cord hematopoietic blood were 55 to 110 fold greater in laserirradiated group than 11 to 27 fold in nonirradiated group. Reincubation of these cord blood cells in the soft agar medium revealed that secondary colonies were formed and the absolute numbers of colony were obviously increased as compared with before liquid culture. The numbers of colony in irradiation group were more than that in nonirradiation group. It suggests that low energy laser irradiation for cord blood stem cells is a effective method in vitro expansion of cord blood stem cells.
