摘要 目的研究甘松挥发油不同途径给药对心肌有效不应期(ERP)的影响,探讨其抗心律失常作用及电生理机制。方法Wistar大鼠50只,体重(250±50)g,分为心外膜局部浸润(局部)(20只)和气道吸入组(吸入)(20只),给予1∶20000甘松挥发油。对照组(10例)。测定心室ERP,ERP/RR比值评价药物的影响。结果①局部吸入组对ERP/RR比值的影响:用药后ERP/RR比值与各相应对照组相比差异有统计学意义(P<0.01),结果与ERP相似。②ERP、ERP/RR比值之间比较:在15、30、45min时间段,用ERP观察比较两种给药方式之间差异无统计学意义(P>0.05),但用ERP/RR比值比较两者,两种给药方式之间差异有统计学意义(P<0.05)。结论①甘松挥发油局部、吸入可明显延长心室肌ERP。②用药后15~45min之间效果明显且稳定,45min后作用明显减弱。③ERP/RR比值能更准确的反应药物对有效不应期的作用。④吸入较局部浸润更加有效。 Objective To investigate the electrophysiological mechanisms of the volatile oil of nardostachys chinensis batal (GanSong) (VOGS) by measuring the Effective Refractory Period (ERP) and value of corrected ERP (ERP/RR) of ventricular myocardium on rats. Methods 50 Wistar rats were used and divided into control group (n=10) and treatment group (n=40)(1:20 000 of VOGS was administrated). The treatment groups were subdivided into local epicardium infiltration group (LEIG) (n=20) and airway inhalation group (AIG) (n=20). Results (1)The effects of LEIG and AIG on ERP/RR. The effects of ERP/RR comparing with control group there are significance difference (P〈0.01). (2) The comparison of effects between the ERP with ERP/RR. In 15,30,45 rain, there are no significance difference in ERP between the LEIG and AIG (P〉0.05), but there are significance difference in ERP/ RR between the LEIG and AIG(P〈0.05). Conclusion (1)The ERP of ventricular myocardium of rats can be remarkably prolonged by VOGS administrated through LEIG or AIG. (2)The effects of ERP was prolonged and steady in 15- 45 min,and gradually was diminished after 45 min. (3)ERP/RR adjusting by the heart rate can really demonstrates the effect of ERP of the myocardium of rats. (4)It shows that administration strategy by AIG is better than that by LEIG.
