摘要
目的构建靶向RhoC基因的RNA干扰(RNAi)载体,并研究RhoC基因沉默对肝癌细胞增殖的影响。方法将合成的寡核苷酸片段克隆入RNAi载体,并转染肝癌细胞,以沉默RhoC基因表达,RT-PCR检测基因抑制效果。绘制生长曲线,检测转染细胞的增殖情况,FCM检测细胞周期变化。结果构建的RNAi载体有效地抑制了肝癌细胞RhoC基因的表达,RhoC基因沉默显著地抑制肿瘤细胞增殖,增殖期细胞百分数显著下降,而静止期细胞百分数显著升高。结论RhoC基因沉默能够抑制肝癌细胞的增殖,为靶向RhoC的肿瘤基因治疗奠定了基础。
Objective To construct the RNAi plasmid targeting RhoC gene and explore the effect of RhoC gene silencing on the proliferation of hepatocellular carcinoma cells(HCC). Melhods Clone the synthetic oligonucleotide fragment into RNAi plasmid, and transfect the constructed recombinant plasmid to HCC for RhoC gene silencing. Determine the gene silencing effect by RT-PCR, the proliferation of transfected HCC by plotting a growth curve, and the cell cycle by FCM. Results The constructed RNAi plasmid inhibited the expression of RhoC gene in HCC effectively. RhoC gene silencing inhibited the proliferation of HCC significantly. The percentage of HCC at G2-S phase decreased significantly, while that at G0-G1 phase increased significantly. Concision RhoC gene silencing inhibits the proliferation of HCC, laying a foundation of tumor gene therapy targeting RhoC.
