摘要
建立了分光光度法测定硫酸皮肤素。在Britton-Robinson缓冲液(pH6.1)中,硫酸皮肤素与吖啶橙形成离子缔合物,染料褪色,硫酸皮肤素浓度与体系在490nm处吸光度的降低(ΔD)成正比,在0.1~3.5μg/ml范围内符合朗伯-比尔定律。定量限为0.1μg/ml。
A spectrophotometric method was established for the determination of dermatan sulfate. In pH 6.1 Britton-Robinson buffer solution, dermatan sulfate binds with acridine orange to fade the dye colour. The dermatan sulfate concentration was directly proportional to the degree of fading (AD) at wavelength of 490nm and obeyed Lambert-Beer's law in the range of 0.1 - 3.5μg/ml. The LLOQ was 0.1 μg/ml.
