摘要
目的:研究大鼠双侧海马立体定向注射Aβ1-40建立阿尔茨海默病动物模型的方法。方法:通过Mor-ris水迷宫试验观察大鼠双侧海马立体定向注射Aβ1-40对其记忆能力的影响;以及改良刚果红染色法检测Aβ1-40注射后大鼠海马组织的病理学变化。结果:大鼠双侧海马立体定向注射Aβ1-40显著导致大鼠空间记忆能力损害,注射点周围有大量的胶质细胞活化。结论:大鼠双侧海马立体定向注射Aβ1-40是一种比较理想的研究炎症在AD发病机理中的动物模型。
Objective To establish the rat model of Alzheimer's disease (AD) by injecting Aβ1 -40 into the bilateral hippocampuses with stereotaxic coordinates. Methods The effect of Aβ1 -40 injection on the memory impairment of rats was evaluated by Morris water maze and the pathologic changes of rat hippoeampuses after Aβ1 -40 injection were observed by improvement congon red staining. Results The injection of Aβ1 -40 into the bilateral hippocampuses with stereotaxic coordinates markedly impaired the spatial memory of rats and significantly activated the glias around the injection points. Conclusion The rat model of Alzheimer's disease injecting Aβ1 -40 into the bilateral hippocampuses with stereotaxic coordinates is an ideal model to study the role of the inflammatory reaction in the pathogenesis of AD.
