摘要 目的:观察血管紧张素-2(Ang-2)对血管内皮细胞[Ca2+]i的影响及合贝爽的保护作用。方法:将血管内皮细胞分为空白对照组、Ang-2组、Ang-2+合贝爽组。Ang-2组加入10-7mol/L Ang-2,Ang-2组+合贝爽组加入10-7mol/LAng-2和1 mg/L合贝爽。各组在药物作用60 min后,用激光共聚焦显微镜测量各组细胞的[Ca2+]i。结果:①Ang-2组的细胞内[Ca2+]i显著高于空白对照组(P<0.01);②Ang-2+合贝爽组的细胞内[Ca2+]i显著低于Ang-2组(P<0.01)。结论:Ang-2可引起细胞内[Ca2+]i的显著增加,而合贝爽对Ang-2所致的[Ca2+]i增加具有保护作用。 Objective: To explore the effects of angiotensin - 2 (Ang - 2) on concentration of intracellular free calcium in vascular endothelial cells(VEC) and protection of herbesser. Methods:Cultured VEC were divided into control group, Ang - 2 group and Ang - 2 +herbesser group. 10^-7mol/L Ang - 2 was added into the culture solution of Ang - 2 group, and 10^-7mol/L Ang - 2 and 1 mg/L herbesser were added into the culture solution of Ang - 2 + herbesser group. After the cultured VEC were treated for 60 min, the concentration of intracellular free calcium of three groups was measured by laser scanning confocal microscopy. Results:The concentration of intracellular free calcium of Ang - 2 group was significantly higher than that of control group (P〈0. 01), the concentration of intracellular free calcium of Ang - 2+herbesser group was significantly lower than that of Ang - 2 group(P〈0. 01). Conclusion: Ang -2 can induce elevation of concentration of the intraeellular free calcium in VEC and herbesser can inhibit the induction of Ang - 2.
